Once again, I'll advise all the people I know who want to install this new OS to back up, back up and back up again...
I personally don't rely on Time Machine for life-saving backups ; I own a license of SuperDuper, which is an excellent, excellent drive cloning utility (did I mention it's excellent?), and I regularly clone my system and data drives onto two separate external hard drives. Yes I know I'm nuts, but at least I'm a safe nut ;)

A few points I see:
-you can't do really make your app work offline on smartphones/tablets
-you can't post pictures
-it's slower because you have to do more calls = more "waiting for reply"
-adding attachements is more painful
-no push notifications (thanks +David A. Fenton)
-it's got to reload the app each time you relaunch the browser (my experience with gmail on safari/ipad)
-video capture (thanks +Nicolas Roberge)

- full text search on a local database is slower because you're going via JavaScript APIs rather than native code
- distribution of content relies on a central point (or CDN) - scaling natively could implement informal mesh networks (have seen this in any apps yet, but would be very useful for certain use-cases)
- integration of camera based augmented reality is not really viable on HTML5
- no image processing
- no audio processing/manipulation
- clunky usability experience to payment gateways (requires a different cognitive load to what users are accustomed to on iTunes. Would argue that Android stores are still very fragmented.)
- no memory management tools for dev/debugging.
